Instrumentation

High Field Mössbauer

The SEE CO. high field Mössbauer spectrometer contains a cryostat with a variable field superconducting magnet, capable of taking data from 1.5K to 300K and 0 to 6T.

Low Field Mössbauer

Three MS4WRC Spectrometers with a 4.5 to 300 K closed-cycle Helium-refrigerated system and a W106 temperature controller. Mössbauer spectroscopy allows us to observe all the iron present in our samples.

UV-Vis Spectrophotometer

Hitachi U-3310 UV-Vis Spectrometer with a Head-On Photomultiplier tube, for performing electron absorption experiments, namely determination of heme centers in isolated mitochondria and measurement of DNA, RNA or protein concentrations.

ICP-MS

Agilent Technologies 7700 Series ICP-MS with ASX-500 Series ICP-MS Autosampler, which allows us to determine the absolute concentrations of different metals in our samples.

Bioreactor

3 Custom-designed 24 L bioreactors from ChemGlass. We need large batches of cell culture to obtain sufficient quantities of isolated organelles for analyses with Mössbauer and EPR spectroscopies.

Ultracentrifuge

Beckman Coulter Optima L-90K Ultracentrifuge with an SW 32 Ti rotor. Used for density gradient centrifugation of organelles to achieve greater purity of isolated organelles.

Anaerobic Glovebox

MBraun LabMaster I30 Glove Box with Siemens Simatic Panel. The glove box has a Nitrogen atmosphere, thus providing an anaerobic environment for isolation of organelles to preserve the oxidation states of iron-containing species in our samples.

Plate Reader

Tecam Infinite M200Pro multimode plate reader. The plate reader measures absorbances, flourescense and luminescence. Multiple wells allows for multiple sample reads at a time and smaller volumes.

Lyophilizer

Labconco FreeZone 2.5 Plus Benchtop Freeze Dry System designed for light sample loads. Used to concentrate the solutions containing low-molecular-mass complexes in our organelle isolations.
