On-target Proteolysis for MALDI-MS
In solution proteolysis requires much time and involves the loss of polypeptides. To decrease digestion times and increase peptide recovery is an important requirement for high-throughput and sensitive proteomic analyses. Optimization of reaction conditions includes determining the effects of organic solvents, temperature and reaction time on the efficiency of rapid on-target proteolysis. This technique can be used for bottom-up proteomic approaches (peptide mass fingerprinting) for MALDI-MS/Imaging-MS experiments.
